dc.description.abstractUna de las estrategias para conocer la función biológica de las proteínas es identificar su localización intracelular. En este sentido las proteínas fluorescentes han sido una excelente herramienta molecular. Metarhizium anisopliaees un hongo entomopatógeno capaz de matar al menos siete órdenes de insectos. Su metabolismo le permite crecer en escases de nutrientes y en presencia de compuestos tóxicos para otros hongos. Sin embargo, el tiempo que tarda en matar al insecto plaga es prolongado, lo cual limita drásticamente sus aplicaciones. Por esta razón se han realizado diversos estudios con el propósito de encontrar posibles nuevos factores de virulencia quepermitan aumentar su capacidad patogénica. Entre estos, en nuestro grupo se han aislado 6 genes que codifican proteínas con actividad de 2-nitropropano dioxigenasa. Con el fin de avanzar en el conocimiento de lafunción de estos genes, propone la localización intracelular de proteínas codificadas por estos genes 2np, mediante fusiones génicas con proteínas fluorescente. En este proyecto se analizó la expresión en E. colide plásmidos que portan distintas proteínas fluorescentes, con el propósito de identificar aquellas que sean másadecuadas para su utilización en el etiquetamiento de los genes 2npes_MX

dc.description.abstractEnglishOne of the main strategies in order to know the biological function of proteins is try to find their intracellular localization. In this sense the fluorescent proteins are one of the most successful molecular tools. Metarhizium anisopliae is an entomopathogenic fungus commonly isolated from soil and insects from all continents, except the Antarctica, and is capable of killing at least seven orders of insects. Metarhizium is capable to grow in a shortage of nutrients and toxic compounds produced by other fungi. However, the time it takes to kill the insect pest is prolonged, which severely limits its applications. For this reason there have been various studies in order to find potential new virulence factors that increase their pathogenicity. Among these, in our group we have been isolated 6 genes encoding proteins with activity of 2-nitropropane dioxygenase. In order to advance the understanding of the function of these genes,we are interested in the intracellular localization of proteins encoded by these 2np genes, using chimeric fusions with genes producing fluorescent proteins. In this project using E. coli as a host we analyzed plasmids carrying different fluorescent proteinsin order to identify those that are most suitable to labeling the 2np genes-
